"Optical Modeling and Fluorescence Spectroscopy Experiments towards Robust Quantification of Biomarkers for Medical purposes"

Abstract

Spectrophotometric analysis has many advantages for in vivo characterization of human biological tissues and classification of healthy or pathological tissues: it is fast and non-invasive. The search for spectral patterns allows for example to detect the presence of biological components, chromophores or fluorophores, characteristic of some tumors. The challenge of the BIOSPEC project is to be able not only to detect but also to quantify these components, i.e. to estimate their concentration in a reliable way. The presented work will give an overview of all activities including experimental aspects in dermatology and neurosurgery concerns as well as improvements of existing light propagation models and their extension to fluorescing tissues. In addition, the presentation will highlight the specificity of biological tissues in an optical point of view: firstly, their high translucency implies a more rigorous description of the light propagation than for more opaque materials; secondly, in vivo measurements imply some constraints (e.g., reflectance measurements are allowed, but not transmittance measurements) which limits the data available for evaluating the parameters of the models.
